The American superstar Adele wanted to be in a whole night on the stage Sunday colossal pyramid.
But, through gritted teeth, cockney singer was forced to reject the offer because she lost her voice.
Instead of Beyonce called his friend Gwyneth Paltrow to take its place.
A source said: "Adele was absolutely gutted, Beyonce is one of his heroes.
"It would have been the stuff of dreams to achieve with it - the concert of his life.
"But docs told her she has to rest his voice or the risk of damaging their valuable asset.
"She hopes one day they can do something together."
This is the second big concert Adele was forced to miss in recent months - albeit for very different reasons.
Prince William and Kate Middleton approached her to play the royal wedding reception, but Adele was unavailable - it was a barbecue reserved.
His battle with laryngitis also led to Adele rescheduling his American tour.
She was told not to sing or even talk for a month or damaging the voice of long duration.
Adele wrote on his site: "There is nothing I can do but listen to the advice and rest."
Beyoncé, meanwhile, has not been easy to take during his stay on Worthy Farm.
She and her husband Jay-Z spent the weekend in Somerset and the couple have not exactly been locked in their hotel room watching Wimbledon.
Gwyneth and the couple were spotted looking COLDPLAY on Saturday.
Beyoncé was also wandering around the site with an assistant, taking in some concerts to wear a mask to protect his identity, according to BBC 6 Music.
It is unlikely that she kept her disguise when she spoke to Bono and Chris Martin ahead of his performance.
Beyonce admitted she caught a few "tips" from other headliners of the festival.
